Yes, I was just wondering.
If I want to have the most accurate estimation of reaction efficiency.
If I already know that I can do a nice standard curve using dilutions of (whatever) 10e1, 10e2, 10e3, 10e4, 10e5. All points lie on line.
I can spare 15 wells for standards.
I could run each dilution in triplicate.
Or I could do 8 of the 10e1 dilution and 7 of the 10e5 dilution.
What would give the best estimation?
I've noticed that the LightCycler 480 doesn't allow the construction of a standard curve unless you have more than two dilutions.

Or, you could do the 10e1 by itself and draw the line through origin. Not.

I'd do the standard curve in triplicate, and samples in duplicates or triplicates.
