Help! Cell staining problem - (Oct/11/2004 )

Hi everyone,

I am now doing cytospin and use safannin, bromopbenol blue to stain cell. When I observed the stained cell slide, there were quite many pink spots or smear between the cells. What is the cause of this? How to solve?? Thanks a lot!

The only way that I know how to stain slides is as follows:

1) Place subject on slide with a small amout of pure water
2) Place cover slip on subject at a 45 degree angle
3) Place a drop of stain near the edge of the cover slip (NOT on the cover slip)
4) Get paper towel and draw fluid out of the side not containing the dye (dab around)
5) The stain will then be drawn into the suject under the slip cover

what I use to stain cell is a thing called cytocentrifuge, it can spin the cell suspension on the slide and soak out the supernatant. The protocol I use says stain the cell first and put cover slip when it dry. And the dye I use is Hemocolor 1 2 3 solution set.

Hope more people to dicuss in the forum!
