Staining phosphoproteins on the SDS gel? - Can Pro-Q Diamond be used for a crude sample? (Sep/13/2008 )

I'm gonna compare protein phosphorylation level ( for all the proteins in the tissue) between two different treatments. I'm wondering if phosphoprotein staining methods like Pro-Q Diamond can be used for this purpose. I've been told that with the crude sample my chance is very small and most probably I'll end up with a smear on the gel for either treatment; cause about one third of proteins are always phosphorylated in some extent. Does anybody have any experience in this regard?

no good experiences with pro Q diamond; better use anti-phospho Y/S/T antibodies in Western
