cell cryopreservation in -80 - (Aug/18/2008 )
hi, there will have any consequences on my peripheral blood mononuclear cells if i transfert them from nitrogen liquid to -80.
please help me because liquid nitrogen is very expensive for my laboratory

I've been keeping my cells in -80 for 6 months now, when I re-grow them they grow very fine. I have no problem with that. my friend keeps them in -20 for 2 months, also he has no problem.
but I'm not sure for transferring from liquid nitrogen to -80, this is a big temperature change.

well the ideal would be to thaw your cells, culture them 1 -2 passage and freeze the culture at-80°.
The heat shock may be too strong.
