Can we store the gel slice after cutting out the band and before gel extraction? - (Jun/30/2008 )
My bands contain 5kb and 2kb size vector and 600 bp inserts. Can I store the gel slice overnight and resume with gel extraction the next day? Will it affect the yield in any way? I was thinking of storing the slice in a 2 ml eppy(autoclaved) and hopefully free of nucleases...at what temp should I keep it --- 4 C or -20C?

I routinely store gel slices at -20C without any problems, sometimes for weeks. Just be sure to weigh the slice before freezing so you know how much buffer to add when you want to purify.

Sure, same as above. -20'C is preferable. You may loose the restriction overhangs or PCR A overhangs over a prolonged storage and freeze-thaw, so cloning strategy may suffer slightly over a period of time, but it has worked for me for months.
