How to know the sequence is in frame - (Feb/05/2008 )
I will cut the insert from pcDNA3.1 and insert it into a shuttle vector, so I have to know the sequence is still in frame. How to check it? I guess some kind of software can do it. Thanks!

Are you creating a fusion protein?
You could use the vector NTI program. Just have the sequence in and then you could translate it and check if its in sequence.

Thank you for your prompt reply, I don't make fusion protein, but a His-tag after the insert, I a beginner of molecular biology, I really appreciate your kindly help.

Some pcDNA have His tag in the vector which makes life more easier. You only have to clone the insert in the sites and you will have his tag at the end and in frame.
