kit for bisulfite treatment - (Feb/01/2008 )

Hello,

I'm going to start DNA methylation analyses by DNA sequencing with bisulfite treatment. I want to know which kit give the best results.

I have seen many kit (Sigma, Zymo, Applied, Qiagen...)

all kits use very similar methods and reagents and are column based.

I have to say though I am aware of only one that protects your DNA throughout the conversion process and that's MethylEasy by Genetic Signatures.

It really depends on your sample, do you have ample amounts? or very scarce amounts to work with?

Thanks for your reply, i have ample amounts and i want to do with the same sample treated "many" fragments, so i hope to find a kit which allow an important amont.
Do you know if you can amplify your treated sample by phy 29 kit (genomiphi for example) in order to have much more matérial?

i hope you can answer and that my english is not so bad!

how many fragments are you planning to amplify?

from 2ug of DNA using a standard homebrew method you can get 50 PCRS from it. so I would say the kits are equivalent.

I am not too sure about phi29, because there will be a base composition skew after bisulfite conversion (all unmeth C's become T) and I am not too sure if phi29 would introduce bias.

If you have ample amounts then there shouldn't be a problem!
