Is it possible to loose the insert? - (Jan/24/2008 )

Hi,

I was given a clone but I l had my doubts because the expression was null to zero so I requested a sequentiation and it turned out that there was NO gene on it. There was no negative clone, so it couldn't be a plaque switch.

My question is, is it frequent to loose the insert?

could you give us some further information?? Are we talking about a plasmid?? A virus??

Some DNA fragments are unstable because of secondary structures and we had a similar incident 5 years ago.
