BAC vecor cloning - (Aug/09/2007 )

i am working to construct a library using BAC vector. can u tell me which host is better to electroporate and clones.

for such size of DNA, electroporation is not recommended. It's better to heat-chock transform.
I would suggest DH10Bac strain. This was the one i used with pFastBac

thaw bacterias on ice. addDNA
heat them 42° 30''
put on ice 2'
lput them on rotating dry-heater 37° 45'
