milk sample in western blot - (Jul/13/2007 )

Hi
I have some problems with my samples.
I try to test Human and Bovin milk in western blot. I haven't problem with the human milk, I see my protein very well. But it's not the case with the bovin milk and i KNOW that this protein exist in .
What can I do ? I denaturat my sample by heating 5 min with DTT. Maybe it's not strong enough?

is your protein known for extensive posttranslational modifications? f.i. casein is multiple phosphorylated; PTMĀ“s may sterically hinder antigen-antibody interactions...
