transformation of some uncommon bacterial - B licheniformis (Apr/26/2007 )
Hi all
is there any common protocol for transofrm plasmid into gram positive bacterial, indeed, I would like to play with bacillus licheniformis but this bacteria do not have some standard protocol for transformation.........
so, I am looking for some protocol that can transform any kind of bacteria......? electroporation ?

I have used a kit from MoBiTec using B. megaterium protoplasts that were made by partially treating the cells with lysozyme which worked quite nicely, if you look in their product manual there are a bunch of references for such a procedure.

Electroporation is going to be the easiest way to go if you don't have a protocol for transformation. You might have to try a range of voltages to find what is the best to use with your cells.
