E. coli lysis with Cellytic and Lysozyme... - (Apr/05/2007 )

I had very nice conditions to lyse Bl21 with cellytic (Sigma) and lysozyme but for some reason sometimes the lysozyme seem to be not active. I had this probleme before and I don't know what to do! the lysozyme is stored in powder at -20C and I prepare a fresh solution 100mg/ml in water. Sometimes it takes 5min to get a clear extract sometimes it doesn't work at all... If someone has a bit of clue.....

could the enzyme batch be bad?

It does happen on occasion. There was a batch of polymerase Long template by Strategene that was bad and there was a world wide recall.

Perhaps your batch of enzyme is bad, or mishandled during transport and has gone bad.
