sepharose bead - (Jan/19/2007 )

If pack a sepharose bead into a column, can that only be used for purification by gravity?

What are you trying to do with it? you can run it with pump.

if it is certified up to which pressure beads are stable you can use it with pressure e.g. HPLC

pump is a kind of pressure, isn't it? It's not flow by gravity, right?

It provides a constant flow rate. The pressure is proportional to the flow rate you apply.

both yes.

If you have a large volume (say 200ml) of sample to load into the sepharose bead column, it is better to use a pump or else you will be staring at the column and adding sample again and again.

If you have a small volume (say <30ml), it is OK to use gravity.

You can use a pump with a flow rate of 1ml/min.
