Knock out in Operon systems - Advice (Oct/03/2006 )
hello. i need some technical advice. my prof wants me to do a knock out of a certain gene in salmonella. this gene is part of an operon complex (structural genes). if ill be doing a knock out of that gene, will it not affect the other genes in the operon? if it will, whats the best way to do it?

I think you should do an in-frame deletion (we call it clean deletion mutant in my lab) of the gene you wanna knock out--which will not affect the other genes in the operon. If you wanna construct an insertional-deletion mutant by throwing in a plasmid to knock out this gene, there is possibility of polar effect.
