This is an old question but here is a possible answer - here is a Mycobacteria Protocols book - available online http://books.google.com/books?id=_4-G0bm5q...6&ct=result

Also, I have a question for anyone who works with Mycobacterium Smegmatis - Is Middlebrook ADC growth supplement (albumin, dextrose, catalase, sodium chloride) required for growth with middlebrook 7H9 broth base? or does it increase growth rate? I've seen at least one paper where they do not state that they add ADC. I'm not completely sure ADC's purpose. Sigma states that ADC supplies energy (glucose) and maintains osmotic equilibrium (NaCl). I plan to test chemical compounds against smegmatis growth (perhaps MICs).

I would enjoy any information about basic culturing of Mycobacterium Smegmatis, as I have no experience and do not know anyone with experience and I would like to pursue some studies involving this bacterium.

Thanks.
