beside triton x-100 we add saponin to permealize cells; 5 min will do; there are humidified chambers to buy but incubate in a light-protected box with high humidity by adding water-wet sponge or cellulose paper
Hi kosmodrom,
Thank you for your suggestion!

The kosmodrom humidified chamber's description is right

In our lab, we use black boxes which present heightenings inside. We add few water and we lay the slides on the heightenings (and we close the box). So you just need a light-protected box with heightenings inside (or water-wet sponge even I've never used this).

As far as humidified chambers are concerned, you can wet a paper towel with ddH20 and line the inside of a tuperware. Place slides inside tuperware, and tuperware inside a 37C incubator. Tends to work for me.
