Can we freeze mitochondria after isolation? - (Aug/10/2006 )
Hi!
Can anybody tell me if we can freez mitochondria after isolation????
I want to do immunostaining on isolated mitochondria next day, so will they be viable next day or not??? though viability is not needed but I want them to stick to the coverslip.
and what if I want to do functional studies on these mitos???

Functional studies shouldnt b a problem after freeze thaw except u wil lose a bit of activity. U have to calculate that for urself.
For immunostaining, just fix them and then leave them in PBS-4C and work on it when u have time.

For fixing, do they need to be in suspension..... I generally put a drop on coverslip, and in 15-20 minutes they stick to it and then I do fixing/immunostaining.
After isolation, I place them on dry ice in a tube... and plan to do fixing and immunostaining next day after thawing. I guess it will work if what you said is right. what you say???
