Fixation protocol for cytoskeleton - (Aug/09/2006 )

Hi,
I am looking for a fixation protocol for the cytoskeleton in neurons. I´ve read that PFA is not ideal for viewing the microtubules and methanol is incompatible with phalloidin staining. Does anyone know a protocol for fixation that would allow me to see both the microtubules and the actin filaments (with phalloidin)?

Try fixing with formaldehyde 4%.

We have used PFA for fixing neurons and visualising tau in the axons.

4% pfa has not caused problems for my microtubules........they are fine. but not neuronal, so maybe that is the difference
