NF-kB staining failed - (Jul/27/2006 )

Hi Dear all,

I have a problem and need your help. rolleyes.gif rolleyes.gif

I have some mouse liver slides need to be stained with NF-kB(p65). Before staining my sample. we did an antibody dilution test with positive control(mouse kidney slides). We chose the ideal antibody dilution used for our mouse liver sample. However, I can not see any signal after staining. I am sure there is no any mistake in the experiment process, and also the liver tissue should have strong NF-kB expression.

Now I feel very depressed, I do not know what I should do to confirm the final results, restain new slides? change the antibody? It is painful..... sad.gif sad.gif

hi,

can you detail a little the staining procedure?
was the sample fixed before storage, and what were the storage conditions?

you should stain some fresh samples to get the best results...
