problem with elisa - (Jul/24/2006 )

dear all,

i have done lowrey's protein estimation and elisa testing on an ammount of protein sample today, and i have observed the ratio of both results , i found that elisa is showing higher result than the low
rey's protein the same amt. of protein, what's the reason? plz tell.

Elisa is a very sensitive method and you didn't give us enough details. But I'll say you must check the background in your Elisa with the negative control if you did n't do that. unsure.gif

cud u tell us
1- how much of estimated protein in lowry's method used for ELISA?
2-how u r comparing the ELISA absorbance reults with estimated protein?
3-do u have any standard for comparing ELISA results?
4-r u doing direct or capture ELISA?
