Plasmid DNA purification - Lithium chlorid (Feb/08/2006 )

Hi!

I am generating real time PCR standard curves with plasmid DNA and it works very well. But I realized that I have a considerable amount of small RNA nuc. in my plasmid miniprep by checking it on a gel.

Now i want to know, if

first: my efficiency is affected by excess RNA?

and second if Lithium chlorid precipitation is appropriate to remove excess RNA, especially small RNA nuc., in my preps?

I did LiCl treatment with 4M LiCl but the RNA didnt disappear form the gel.

Thanks for any comments, SilAttached Image First 2 traces with LiCl treatm.

do rnase treat with phenol chlo or do a CsCl purification if you want ultra pure prep
