vortexing after phenol-chloroform-IMA purification - (Jul/17/2009 )

hey,

is it recommended to vortex the sample after phenol-chloroform-isoamylalcohol addition during a plasmid prep? does it shear the DNA?

The protocol says to mix well as far as I can recall. I just shake the tubes hard for 15 sec.

I guess I wouldn't vortex my sample. The longer the DNA, the easier it is to shear it. I guess I will just invert my tubes for about 15 times.

I usually vortex after P:C:I addition and have had no troubles with pDNA or gDNA downstream.
