probe size for sonicator - (Mar/30/2009 )

we have an ultrasonicator with microtip in the lab. i tried to break E.coli cell down with it. but it did not work. can anyone tell me what is the probe size for E.coli cell lysis? i have no idea which one i should order.

it depends on the volume you wish to sonicate.

check the book that came with the sonicator (or the catalog of the company from which you purchased it). it should tell you what size probe (or probe) to use for which volume.

E.coli should not be to hard to break using ultrasonics. Have you tried to apply mild pressure (.5 to 1.5barg) during sonication using a pressurizable sonication reactor. Amplitude at the tip should be between 30 an 50 micron. I would recommend using a setup comparable to our 500 watts ultrasonic processor UP500hd.
