Tissue & Cell Fixation solutions - (Mar/28/2009 )

Hi,

I am currently seeking a fixation solution for tissue or cells immunostaining, which will instead of use the peraformaldehyde (PFA), does any experts could tell me any information about this?

I've always used PFA with good results.
Why do you want not to use it?
Need more info in order to try to help

I usually use -20 degree methanol for 10-15 minutes rather than PFA. It maintains certain cellular structures better and doesn't require a permiabilization step.

I always use a 4% FORMALDEHYDE solution for 20 minutes on ice, which works well for all my stainings. The methanol methode is also used, it is suppost to work better with certain stainings
