DNA extration of pieces of human hearts - (Mar/18/2009 )

Hey guys,
We differentiate stem cells into cardiomyocites and now we want to study the methylation profile of some genes during differentiation. I have some pieces of human heart and I would like to use it as positive control of DNA methylation.
Which protocol of DNA extraction do you guys suggests in which DNA is suitable to BSP studies?
We have huge pieces of heart like half ventricule or atrium. They are frozen at liquid nitrogen.

Any method will do. You only need a few micrograms of DNA for methylation study. Make sure the extraction method gives you pure DNA.

Here we have a protocol that uses phenolcloroform, this DNA is good for PCR amplification, I was just worried about the enzimatic efficiency of DNA conversion in the presence of such reagent.
