Colour of PCR gel band best for ImageJ analysis - (Mar/27/2015 )

Hello all,

I've just started doing PCR.

Since I can image my gel so that the  bands show up as black or white, which colour is best for ImageJ measurement? It seems to me that white bands contrast better against the background and thus give me a sharper peak in the readings? Does anyone have particular tips on this?

Either should work fine, as they are functionally equivalent in mathematical terms. The eye's perception has nothing to do with actual analysis in terms of binary data sets.
