questions on transwell assay - (Mar/07/2014 )

Hi, I am facing the problems when I do transwell assay to detect cell migration. But the cells looks very "ugly" when i take pictures. Does anyone know how to keep the cell pretty? I wash the transwell with PBS, did I wash it too much? 

Can you add a pic of these "ugly" cells?  At which point are they "ugly" - before or after migration?

After migration. They look "broken" 

What sort of broken?  apoptotic? senescent? ragged? vacuolated? change of shape?

What cells are these?  Is is possible that they are differentiating?
