Disaggregation of micro-algae - (Nov/14/2011 )

Hi

I am currently helping one of our phd students with the development of a bio assay, we need to disaggregate some micro-algae into single cells but wish to keep the cells intact, plants are not an area of expertise for me, u have tried mechanical and ultrasonic disruption but these have had limited success.

Can anyone suggest a method for disaggregating the cells ?

Which methods of mechanical disruption did you use?

If you know what is linking them (cellulose?) you could perhaps use an enzyme digest.
