Signal Peptides - To leave or not to leave (May/27/2011 )
Hi there
I am trying to order some synthetic genes for a protein. The native sequence from the sequence database has a signal peptide and is prokaryotic (i.e. from Bacteria). I have not made any changes to the gene sequence whatsoever.
Should the signal peptide sequences should be removed? Or left?
I am worried about this. Will this express my proteins it in the wrong place in the cell if it has no signal?

Hola with the signal peptide your protein is exported and processed in periplasmic space, without it protein is expressed intracitoplasmic. For purification you have to do an osmotic shock in order to have a quasi pure protein fraction, but if you donīt made and sonicate or broke bacteria you will have a mix of non processed intracitoplasmic sp-protein and procesed periplasmic protein. buena suerte
