Ct value and interpretation - (Jan/06/2011 )

Hi
I am comparing the gene expression levels between two groups (Treated and untreated).
How do I present the results? In terms of Delta Ct values or Ct values normalized to housekeeping gene?
Please advice! I Dont think I can use delta delta Ct since I have only 2 groups and I may not be able to plot a bar graph :(
I appreciate your suggestions

use the control and substrate the treated values from control. assume control samples as 1 and any change is related to this control.

is this helpful?

Hi Scolix
Thanks for the advice. I am going to use this now :) THanks for your time!

Hi-

You need to normalize the Cts to the endogenous control- get the DCt and then compare the control to the target group for fold change. You can calculate the DDCt with two samples.

Take a look at this PCR whitepaper http://www.integromics.com/WhitePaper_qPCR_Statistics

and also the Livak paper.
