What's the solvent I should use to dilute my organic compounds - compounds dissolved in DMSO, but cannot dissolve in aqueous solution (Jun/19/2010 )

Hi~~ I'm trying to analyze how my sythetic compounds will bind to a specific protein using Elisa. I have stock compounds in DMSO (10nM). But now I need to dilute them for the elisa assay. What's the solvent I should used? I understand that the concentration of DMSO should not higher than 0.1%. (??) I try PBS but they just percipitate....Any suggestion for that? Thanks!!

Try to solubilize your compounds in aqueous buffer rather than DMSO or use the least concentration DMSO possible. Might there be another organic such as ethanol that could be used?

You may also try adding a surfactant.

The concern is having the solvent and/or surfactant in the assay at a concentration which would be detrimental to the performance of the test.

Hi~ What kind of surfactant do you usually use?

Try to solubilize in Triton x -100 or Tween-20. You may wish to run a control if you have one with the same level of surfactant to determine any negative impact upon the test.
