Hi guys,
Quick question. What purpose does the little white disk (frit) serve for a protein A chromatography column? My co worker says he never uses them and that its unnecessary. Is that true?

it is used to prevent flow through of column matrix material...

That would be the bottom frit. Top frit is used to prevent gel disturbance by the flow (sample application, wash/elution/whatever buffer) which may have negative influence on separation.
