Fluorescence-black plates? - (Feb/04/2010 )

Dear all,
I am going to measure flluorescense of AMCA, but I have only black plates with transparent bottom. Can I cover the bottom (with adhesive sheet) and still measure fuorescense or I can use ONLY whole black plates with black bottom?

You could cover the bottom, but you have to be careful about what material you choose. Many materials are fluorescent, and you have to avoid those. A blank would reveal excessive fluorescence in the blocking material. It's possible that simply using the plates as-is would be the best solution. I think the inside of your plate reader is likely to be quite dark. There may be extra well-well bleeding of light, but I would guess that to be minimal.
